Abstract:
Background: Endothelial dysfunction considered as an initial and developing event in the progression of atherosclerosis in patients with metabolic syndrome. Piceatannol have cardioprotective effects and anti-inflammatory properties in several diseases. In the present study, we investigated whether piceatannol provides beneficial effects on hyperglycemia /hyperlipidemia-induced dysfunction of endothelial cells and explored the possible molecular mechanisms. Methods: Human umbilical vein endothelial cells (HUVECs) were exposed for 72h to high-glucose/high lipid (HG/HL) using dextrose (33 mmol/L) and the saturated free fatty acid palmitate(500 mmol/L) , with or without pre-treated of piceatannol (10uM) for 24h. HUVEC viability, proliferation and migration were determined. Protein expression of endothelial intercellular adhesion molecules (ICAM-1), endothelial nitric oxide synthase (eNOS) were measured by Western blot and some key signalling pathways that regulate endothelial cell function were also characterized.. Results: We found that hyperglycemia and hyperlipidemia resulted in significantly impaired endothelial cells function as shown by the reduction of HUVEC viability, proliferation, cell migration and the protein level of the phospho-endothelial nitric oxide synthase (p-eNOS). Dextrose and the saturated free fatty acid palmitate stimulation of HUVECs increased the level of the Intracellular adhesion molecule 1 (ICAM-1) and accelerated endothelial senescence. These effects of hyperglycemia and hyperlipidemia were inhibited by pretreatment with piceatannol for 24h. These beneficial effects of piceatannol on the endothelium were associated with MAPK and NF-KB pathways. Conclusion: Piceatannol, via increasing the protein level of the phospho-endothelial nitric oxide synthase (p-eNOS), reserving the function of cell viability, proliferation, cell migration, decreasing endothelial senescence, down-regulating the level of the adhesion molecules (ICAM-1), suggests that picetannol exerts beneficial effects on hyperglycemia and hyperlipermia induced endothelial cell dysfunction through regulation of the MAPK families and NF-KB pathway.
